detection of medically important candida species by absolute quantitation real-time polymerase chain reaction
Authors
abstract
results all eight candida species were successfully detected, identified and quantitated based on the icl gene. a seven-log range of the gene copy number and a minimum detection limit of 103 copies were achieved. conclusions a one-tube absolute quantification real-time pcr that differentiates medically important candida species via individual unique melting temperature was achieved. analytical sensitivity and specificity were not compromised. materials and methods the isocitrate lyase (icl) gene which is not found in mammals was chosen as the target gene of real-time pcr. absolute quantitation of the gene copy number was achieved by constructing the plasmid containing the icl gene which is used to generate standard curve. twenty fungal species, two bacterial species and human dna were tested to check the specificity of the detection method. objectives this study aims to develop a method for the detection, identification and quantitation of medically important candida species through quantitative polymerase chain reaction (qpcr). background the number of invasive candidiasis cases has risen especially with an increase in the number of immunosuppressed and immunocom promised patients. the early detection of candida species which is specific and sensitive is important in determining the correct administration of antifungal drugs to patients.

full textDetection of Nocardia Asteroides Complex in Clinical Isolates by Real-Time Polymerase Chain Reaction
Background and Aims: Nocardia asteroides complex is the most common cause of infectious diseases due to nocardiosis. Interspecies differentiation of Nocardia genera is essential for prognosis and timely proper treatment, as well as for epidemiological studies. Since each genus has its own antibiotic resistance, precise careful diagnosis is of prime importance. As compared to biochemical and phe...
full textDetection of castor contamination by real-time polymerase chain reaction.
Due to the potential for intentional contamination of food with crude preparations containing ricin, a real-time PCR method was developed for the detection of castor plant material in ground beef. One primer pair was identified and confirmed to be castor-specific and efficient for amplification of ricin in DNA extracts from castor or beef matrices. Of three different DNA extraction protocols co...
full textDetection of a wide range of medically important fungi by the polymerase chain reaction.
A polymerase chain reaction (PCR) method was developed that was capable of detecting a wide range of medically important fungi from clinical specimens. The primer pair was designed in conserved sequences of 18S-ribosomal RNA genes shared by most fungi. The lower limit of detection of this PCR technique was 1 pg of Candida albicans genomic DNA by ethidium bromide staining and 100 fg after Southe...
full textReal-time polymerase chain reaction.
Real-time PCR is the state-of-the-art technique to quantify nucleic acids for mutation detection, genotyping and chimerism analysis. Since its development in the 1990s, many different assay formats have been developed and the number of real-time PCR machines of different design is continuously increasing. This review provides a survey of the instruments and assay formats available and discusses...
